Overview
Analyzing the cellular behavior at a single-cell level is of critical importance to better understand the heterogeneity of cellular response such as protein secretion or enzymatic activity.
To this purpose Fluorescence-Activated Cell Sorting (FACS) appears as a convenient tool to indicate the production of a molecule of interest. On the other side microfluidics technology and more exactly encapsulation of cells in aqueous droplets allow to study at single cell level the cellular response such as protein secretion or enzymatic.
The combination of these two different technologies offers lot of potential to study single cell expression however a single water in-oil (W/O) emulsion is actually not well suited to FACS sorting.
Indeed, FACS sorting requires that the particles are suspended in a aqueous phase to efficiently sort the particles of interests by applying an electric charged. To avoid these limitations, the W/O droplet can be encapsulated in a 2nd aqueous layer to form a water-in-oil-in-water (W/O/W) droplet, which is compatible with FACS.
In this workshop we will present you how to encapsulate biological materials such as cell inside double emulsions, in an easy and robust way, which are totally compatible with FACS sorting

Our Encapsulation Platform for FACS
The cell encapsulation platform includes an organized flow path with pressure controllers, filters, flowmeters, and valves to facilitate the start-up, shut-down, and cleaning of the system between runs. Dedicated optics are included to optimize the visualization of droplet production at high generation frequency.
How to encapsulate bacteria in double emulsions?
In this application note, we associate the double emulsion microfluidic platform with FACS sorting technology to sort out small droplets encapsulating fluorescent e-coli bacteria.
